5 descendants of David Maple Sr. have done advanced Y-DNA SNP (single nucleotide polymorphism) testing at Family Tree DNA. These all originated sometime after the birth of the nearest common patrilineal ancestor of Benjamin Maple Jr and the Mapley families that lived in Buckinghamshire, England in the 18th century. Family Tree DNA estimates that this common ancestor was born about 1486 CE (95% confidence interval: 1267-1651 CE). Autosomal DNA

The amount of autosomal DNA inherited from any particular ancestor falls off rapidly with each successive generation, so it is difficult to use autosomal DNA to prove descent of currently living people from ancestors as distant as Benjamin Maple Sr, his children, or his grandchildren. Here, we give examples where triangulated DNA matches provide evidence of descent from early Maple generations.

Descent from Jasper Newton Maple (and his great-great grandparents Benjamin Maple Jr and wife) and Jasper's wife Lydia (Chambers) Maple
Figure 1 illustrates some chromosome 14 DNA matches for Bruce Maple within the Family Tree DNA database. The segments marked in red represent DNA (46 cM on 2 segments) shared with a 3rd cousin (once removed) whose nearest common ancestors with Bruce are Jasper Maple and Lydia (Chambers) Maple. A 9.6 cM section of this DNA (cyan) is also shared by a descendant of Mary (Maple) Roseberry, a daughter of Jasper's great-great grandfather Benjamin Maple Jr. Adjacent to the largest segment shared with Bruce's 3rd cousin is a 13 cM segment (black) shared with a 5th cousin who is a descendant of William B. Maple, a son of Jasper's grandfather David R Maple.
A 24 cM segment (orange) is shared with a descendant of Francis Chumard, who appears to have been an uncle of Lydia (Chambers) Maple's grandfather John Chumard.
A 9.1 cM segment (blue) partially overlapping the large segment Bruce shares with his 3rd cousin is shared with a descendant of Dorcas Mayhorn (1794-1860), who lived in Fayette County, Pennsylvania. Bruce also matches another of Dorcas' descendants (on a different chromosome) through a different son. It appears that Jasper's mother, Hannah (Mahon) Maple, and his uncle, Ezekiel Mahorn may have been part of a related family.

Descent from David Maple Sr and his wife Mary (Johnson) Maple
Figure A shows chromosome 9 matches within the Family Tree DNA database for a Maple who is a 3rd great grandson of David Maple Sr's son John Maple.
This tester shares a 21.36 cM DNA segment on chromosome 9 (base pairs 116724402 - 133054779) with a 4th great grandson of David's son Jacob Maple (D).
He also shares most of this segment with 3 descendants of David's son David R. Maple. [A 2nd great grandson of William B. Maple's daughter Ann (Maple) Fuller (B) matches on base pairs 115788707 - 127642231 (15.77 cM), and a MAPLE who is a 3rd great grandson of William's son James B. Maple (C) matches on base pairs 122547228 - 132048664 (9.64 cM). A third descendant of David R. Maple, a 3rd great grandson of both William B. Maple and Joseph Maple (A), matches on base pairs 119878770 - 139720795 (35.74 cM)].
It appears that all of these segments were passed down from David Maple Sr and/or his wife, Mary.
