Denis was born in 1856 in Clooneybreen, Rineen in County Clare, Ireland, the son of Denis O'Dwyer and Anne 'Nancy' Hurley. When he was 22 years old, he married Anne Marrinan in 1878 in Ennistimon in County Clare. Anne joined him at Rineen where they farmed and had ten children together, three of whom died young. Anne died in 1898, and the younger children were taken into the home of their older sibling Catherine. Denis remarried in 1901 to the widow Bridget O'Connor née Comyn. Bridget was a successful business woman running a public house and grocery in Cloonadrum in County Clare. Denis went to live there with her and two of her children from her first marriage, and to farm. They did not have children together. Denis passed away at age 88 in 1944 in Cragaknock, County Clare.
