William was the son of Christopher Kelke and Isabel Gyrlington.[1][2][3] He may have been born at Barnetby le Wold, Lincolnshire where his father had his main property, but his father also had lands in Yorkshire.[4][5] He was born after 3 April 1519, when his father's Will was dated.[6]
William married Thomasine Skerne, daughter of Percival Skerne, a London fishmonger, and Joan.[2][3] They had two children:
William had property at Barnetby le Wold, Lincolnshire. He was also a London mercer.[2][3]
In 1551 William was sued for debts totalling £1031. The official record describes him as a mercer of London.[7]
William's Will, dated 16 June 1552, was proved on 28 June 1552.[2][3][8] It was witnessed by his father-in-law and named his wife Thomasine.[9]
His wife survived him, dying a little before 31 August 1566, when administration of William's estate was granted to Jeffrey Ducket until her daughters came of age.[2][3]
