I'm trying to find a source for Humphrey Booth's title of Captain, whether he was actually in the Colonial Forces or the title was an honorific.  Any clues would be appreciated.

Nancy, is he already in WikiTree? I don't see him. We first need to identify with some certainty which Humphrey Brooke you are asking about. Do you know his father or place of origin? Offspring? Where did he live? I ask because there are a fair number of men of this name over quite a span of time including a Lt Humphrey Brooke but his commission date is 1756.
In WikiTree he is listed as Humphrey Brooke, Sr. 1635- bef 1685.  Father Robert Booth, Mother Frances Burgess, Wife, Margaret Underwood, Daughter, Catharine Booth, m. Robert Brooke.  Born England, came to VA at age 19.  Seems likely he would have been in the VA Militia.  Called Captain in Genealogies of VA Families, Vol 1, p 472 & VA Magazine of Hist. & Bio, Vol 14, p 325-326. I can find no proof of his military service.
Is this a profile you have just created? I can't seem to get him to come up in a search. Can you edit your question and below the question box where it asks "WikiTree ID/URL if this relates to a person or free-space profile" copy the URL (browser address) for his profile? Thanks.

Also it would help to type in Virginia where it asks for Surnames/Tags. That will get the active Virginia eyes on the question for a group assist. You can also add to tags southern_colonies which is now active with several teams of researchers.
I can't find him in a search either.  To get to him,  I put in the surname of Booth, and alphabetized the list, then went down to the H's and found Humphrey Booth, Sr (1635-bef. 1685).  It is not a profile I created.  URL is https://www.wikitree.com/wiki/booth-1033.  As you can tell, I don't know my way around the platform very well. Now I know what a Surnames/Tags is all about.
OK, so the surname is Booth rather than Brooke? I'll look around some more in the morning--can probably find something. No problem on learning WikiTree--it has a steep learning curve but it is well worth it in my experience.

Nancy, I don't yet have a definitive answer for you but this information may aid your search.

I don't find him with any title, other than Gent, through his marriage date 1663. Side note: I see that the date of marriage appears to be 1660 on the wife's profile but most records I am seeing are showing 1663. You might want to source and correct that for both profiles.

Later in life I do not find him given as a Capt Booth with one exception so far found. In a transcription of Virginia Land Book, Richmond, Book VI, 539, "September 1672. Nicholas Catlett and William Moseley, 1,200 acres, adjoining Capt Humphrey Booth, south side of Rappahannock." This is a transcription so my suggestion now is to see if you can find a different transcription (Library of Virginia online is the place to start) and see if it also reads Capt. If it does not, then I think it is likely an error. If it does read Capt then he became Capt of something between marriage in 1663 and this land record in 1672 which narrows the search window considerably. I see no reference to him as a mariner which leaves it to be a military rank if he was a Capt.

Also, go back to your question and click the Edit button. Then enter both Virginia and southern-colonies in the tags, then save it. That will bring this question before the Virginia team and the new Southern Colonies team focusing on Virginia.

It's very common in early Virginia records to find men listed as Captain or Colonel.  It was more of a title showing the person had some position of authority rather than a military title. It may have meant that the person was responsible for creating a militia in his community if needed.
