Samuel was born in 1815 to parents John Lyon and Sarah (Griswold) Gardiner.
He married Mary Gardiner Thompson in 1837.
Samuel was educated in Clinton Academy, East Hampton; Union Hall, Jamaica, Long Island; and Dwight Seminary, New Haven. He preferred a rural to a professional life and lived, both before and after his proprietorship of the Island, the life of a prosperous country gentleman and farmer.
He became the 10th proprietor and "Lord of the Manor" on Gardner's Island upon the death of his brother John Griswold in 1861 and by purchasing the rights of his remaining sister, Sarah.
He took an active interest in the affairs of his native town and state, serving at different times as town clerk, justice of the peace, and supervisor in the town of East Hampton, and as a member of the New York State legislature in 1864 and again in 1876.
The Island was a happy place under the proprietorship of Squire Gardiner. He devoted most of his time to the improvement and management of it, and it prospered as in the days of John Lyon. At his death in 1882, Judge Henry Hedges said of him,
“As a boy I knew him, high-souled, sympathetic, social, generous, friendly, inheriting the activities of his father and nobility of nature that signalized his mother; he was one of nature’s noblemen.”
He passed away in 1882 leaving the ownership of Gardiner's Island to his eldest son, David Johnson who abrogated his rights to his younger brother John Lyon.
