Robert was born on 28 August 1834 at Edinburgh, Midlothian, Scotland. He was the son of Sir Ralph Abercromby Anstruther of Balcaskie, 4th Bt and Mary Jane Torrens.

He was educated at Harrow School, Harrow on the Hill, London, England. He gained the rank of officer in 1853 in the service of the Grenadier Guards.

He married Louisa Maria Chowne Marshall, daughter of Reverend William Knox Marshall and Louisa Marsh, on 29 July 1857 at Beckenham, Kent, England. They had children:

• Mary Evelyn Anstruther 1859-1921
• Henry Torrens Anstruther 1860-1926
• Admiral Robert Hamilton Anstruther 1862-1938 m. Edith Flora Peel
• Arthur Wellesley Anstruther CB 1864-1938 of the Board of Agriculture and Fisheries, m. Louisa Adele Rose Trapman

He gained the rank of Lieutenant-Colonel in 1861.

On the passing of his father in 1863, he succeeded to the title of 5th Baronet Anstruther, of Wrae, co. Linlithgow established 1694 in the Scottish Baronetage of Nova Scotia, and from 1698, "of Balcaskie, co. Fife" and "of Braemore, co. Caithness."

He held the office of Member of Parliament (M.P.) for Fifeshire between 1864 and 1880. He held the office of Lieutenant of Fifeshire between 1864 and 1886.1 He held the office of Member of Parliament (M.P.) for St. Andrew's Burghs between 1885 and 1886.

Sir Robert inherited landholdings in Caithness, particularly in the parish of Watten.

He passed away on 21 July 1886 at age 51.
