Mori Mutsuko was born in Chiba-ken on 31 July 1917, the daughter of Mori Nobuteru, a businessman of the Mori Konzern who would later serve as a member of the Japanese House of Representatives from 1924 to 1932, and Yamaguchi Inu. She graduated from Tokyo Prefectural Daiichi Girls' High School.
Mutsuko married Miki Takeo in June 1940; they had three surviving children, including a daughter named Kiseko, during their marriage. Mutsuko supported her husband's political activities until his death in 1988, and was the spouse of the Prime Minister of Japan from 1974 to 1976.
Mutsuko also supported liberal causes, particularly regarding pacifism, relations between the Democratic People's Republic of Korea (North Korea) and Japan, and compensations for comfort women.
Mutsuko died from colon cancer at a hospital in Tōkyō-to on 31 July 2012.
