Jacob was the son of Gideon Markle and Anna Hart. When Jacob Markle was born on December 29, 1845, in Indiana, his father, Gideon, was 43 and his mother, Anna, was 37. Along with his siblings, John and Amos, participated in the Civil War. "Jacob Markel enlisted in Company I, 57th Regiment, Indiana Volunteer Infantry, October 1861, for three years, was in the battles of Stone River, Chicamauga; veteranized in the same company and regiment, February 1864; was in the battles of Franklin and Nashville, and "is now in the service, May, 1864. Son of Gideon Markel, Dalton tp."

He married Ellizabeth Billheimer on June 23, 1882. They had two children during their marriage. He died on April 15, 1923, in Richmond, Indiana, at the age of 77, and was buried in the Pioneer Friends Cemetery in Webster, Indiana.

Obituary (The Richmond Item, April 18, 1923): "Jacob Markle died at Richmond Saturday evening at the home of his son, Prof. Millard Markle. Mr. Markle was a former resident of Webster. He is survived by one daughter, Mrs. Anna Williams of Webster, one son, Prof. Millard Markle, of Earlham College and one granddaughter, Miss Mildred Williams of Webster."
