James (Hamilton) Hamilton Ist of Stonehouse is a member of Clan Hamilton.
James Hamilton of Stanehouse[1] the first of this family (sometimes called "Sir James") was the eldest son of James Hamilton of Raploch (1503 to 1531) by Isobell Weir his wife.[2][3] In the ordinary course he succeeded to the Raploch estates but, having become possessed of the lands of Stonehouse in right of his first wife Margaret Mowat, he resigned the lands of Raploch in favour of his younger brothers successively, namely, Thomas HamiltonArchibald Hamilton, and Gavin Hamilton, and their heirs-male.[4] The first two of these had no legitimate heirs so the land and title ultimately passed to Gavin.
He married firstly, Margaret Mouat, daughter and heiress of Alexander Mouat of Stanehouse, Lanark, before 27 Nov 1521 when he resigned his interest in Raploch to his younger brothers.[4][5][6][7] They had issue:

He married secondly, bef. 10 Mar 1539/40, Grizel, daughter of Robert Sempill, third Lord Sempill whom he later divorced for adultery.[5][7][13] They had issue:[9][14][15]
He commanded the troops under the Earl of Arran and Cardinal Beaton at the taking of the martyr George Wishart.[19]
He was Captain of Castle of Edinburgh and Provost of Edinburgh from 1544 to his death in 1548[16] and escaped from ward 1544[20] (Criminal Trials). He was killed with his eldest son James quelling a riot between French troops and locals on 1 Oct 1548.[21][22]

As for the confusion concerning the Hamilton Y-DNA and the Douglas Y-DNA. Also the refusal of some to recognise the father of James "5th Laird of Cadzow" Hamilton (Hamilton-1093) as being Sir John Hamilton, 4th Baron of Cadzow (Hamilton-1087) who potentially is my 15th (2X),16th,19th (2X) great-grandfather.
In one instance of the Douglas Y-DNA: Y-DNA I-L338 for all of Walter Hamilton of Darngaber's male line? Possibly. (my early Great-grandfather, on my grandMother's side) Y-DNA is possibly I-Y6635 per Descendent of Walter Hamilton of Darngaber, (potentially one of my FTDNA matches). BOTH I-L338 (also known as I1a2a1a1a) and related I-Y6635 (also known as I1a3a1a1a1, Parent Branch: I-Y6624) are of Haplogroup I-M253 also known as I1 (a Y chromosome haplogroup). I-L338 is a well-known Haplogroup of the Douglases.
