Stephen Hales was the son of a Hales (John or Thomas) of Canterbury, whose brother was John I Hales (died 1572), of Coventry, Warwickshire and London. He married twice:[1]
Amy Amice (Morison) Beale, widow of Robert Beale (died 1548), and daughter of Thomas Morison (abt.1480-) of Cherdwell, Buckinghamshire. History of Parliament says that possibly all of his four sons and at least one daughter were by this first marriage:[1]

Secondly, before 1561, he married Bridget (Over) Nethermill, widow of John Nethermill[2], and daughter of Henry Over.[1]

His will was written 26 Jun 1574.[3][4]Daughter Anne from his first marriage appears not to have been named in his will, which is not uncommon when children are born from a prior marriage.
